 What are a pH Meter and the way does it Work?

A pH meter is a device used to measure the acidity or alkalinity of a liquid solution - also called pH. pH is the unit that describes the degree of acidity or alkalinity. It's measured on a scale of 0 to 14.A pH meter consists of three different parts: an internal electrode, a reference electrode, and a high input impedance meter. 

A glass probe often contains 2 electrodes -- an internal electrode and a reference electrode. The interior electrode may be a silver wire covered with chloride (Ag/AgCl wire), and the reference electrode is usually made from equivalent materials. Inside the probe may be a solution at a pH of seven. Measured pH is the difference in [H+] between the reference buffer inside the probe and therefore the sample solution. The pH measurements are made by comparing the pH reading of a sample solution thereto of a reference solution with defined pH, like buffers. Therefore, it's important to calibrate the instrument with appropriate buffer solutions before making any measurements.

What is Centrifugation?

pH Measurement

pH measurements are typically made employing a calibrated pH meter at 25 °C (or other specified temperature) and provide a measure of the acidity or basicity of a solution. The pH stability profile of the active ingredient within the formulation, combined with the safe pH range for the merchandise in use, will define the suitable limits for the formulation. pH measurements are often taken on the majority product (to assess the extent of any adjustments to be made before completion of blending and commencement of filling), on the finished product (to determine compliance with set limits and release of the batch), and on storage (to give a sign of formulation stability).




pH Electrodes

The combination of pH electrodes measures the difference in potentials between the 2 sides within the glass electrode. to measure the potentials, it must be an electrical circuit. The circuit is closed containing the inner solutions of the electrode and therefore the external solution that's being measured and the pH meter.

As the electrode is immersed within the test solution the glass bulb senses the hydrogen ions as a millivolt (mV) due to the charge of the hydrogen ions. The electrolyte picks up the mV signal from the glass bulb. That signal is then passed to the interior electrode. The Ag/AgCl wire then passes that signal to the electrode cable that results in the meter. The reference electrode containing electrolyte or internal solution produces a continuing mV, which is transferred to the Ag/AgCl wire. The wire helps to transmit the signal, which is often considered a control being measured to the electrode's cable.

Calibration

The pH meter should be calibrated before each measurement using a minimum of two standard buffer solutions of known pH values (typically around 4 and 7).
